泛素化相关通路

摘要:泛素是在细胞质、细胞核和真核细胞中发现的高度保守的小分子球状蛋白。泛素化是指泛素-蛋白质偶联过程。泛素通常通过不同的长度、结合位点和拓扑结构与蛋白质结合,这导致蛋白质翻译后修饰的多样性。这个过程对蛋白质稳定性、亚细胞定位和与其他蛋白质的相互作用有影响。目前,泛素化参与大多数细胞过程、信号通路和疾病调节。

1. 泛素介导的自噬途径

巨自噬是细胞通过溶酶体降解长寿命蛋白质、细胞器和一些细菌的分解代谢过程。自噬途径需要吞噬体的发育。吞噬体包裹细胞质成分形成双膜自噬体,然后与溶酶体融合以消化内容物。自噬通路在预防许多疾病方面起着重要作用,例如神经变性、肌病、肝脏疾病和糖尿病等。据报道,自噬似乎可以促进和抑制癌症。

2. Beclin 1自噬

自噬是由PI(3)富集区域的吞噬体形成引发的。吞噬体也称为欧米茄体。PI(3)PBeclin 34复合物的催化亚基hVps1产生,是募集下游自噬组分(例如WIPI1/2DFCP1)所必需的。ULK1/2激酶复合物(例如ULK1ULK2ATG13RB1CC1)也是自噬起始所必需的,并且还通过促进Beclin 1复合物从微管中释放来正调控Beclin 1复合物。通过磷酸化Beclin 1和刺激hVps34活性可以直接调节。

3. LC3自噬

吞噬体的延伸需要mATG9和泛素样蛋白LC3mATG9主要以跨膜蛋白的形式退出高尔基体网络,并以ulk1依赖性方式与吞噬细胞短暂相互作用。这种相互作用可能有助于吞噬细胞延伸过程中的膜转运。LC3通过两步转换为LC3-IILC3-II是一种常见的自噬标志物。LC3ATG4B切割形成LC3-I。然后,LC3-I通过泛素样活化酶ATG7和泛素样结合酶ATG3转运到吞噬体,并与磷脂酰乙醇胺(PE)结合形成LC3-IILC3-II的形成受到效应蛋白PI(3)P(例如WIPI1/2)和泛素样复合物ATG16L-ATG5-ATG12的正向调节。RB1CC1可能与ATG16L相互作用。ULK1/2激酶复合物可能在吞噬细胞的延伸中发挥作用。LC3-II与底物结合,通过不同的衔接蛋白进行自噬降解。在线粒体自噬过程中,LC3-II通过BNIP3L衔接蛋白与线粒体结合。

4. 溶酶体中的自噬

自噬体由双膜自噬体形成。但机制尚不清楚。GATE-16LC3密切相关,只是自噬体成熟后需要的。它可能在膜的融合中发挥作用。当自噬体与溶酶体融合时,自噬体的降解导致膜损伤和内容物消化。
